Validation of novel hub genes and molecular mechanisms in acute lung injury using an integrative bioinformatics approach

Acute lung injury (ALI) is an inflammatory pulmonary disease that can easily develop into serious acute respiratory distress syndrome, which has high morbidity and mortality. However, the molecular mechanism of ALI remains unclear, few biomarkers for diagnosis treatment have been identified. In this study, we aimed to identify novel using a bioinformatics approach. Gene expression data were obtained from Expression Omnibus database, co-expressed differentially expressed genes (CoDEGs) identified R software, further functional enrichment analyses conducted online tool Database Annotation, Visualization, Integrated Discovery. A protein–protein interaction network was established STRING database Cytoscape software. Lipopolysaccharide (LPS)-induced mouse model constructed verified. The hub screened validated in vivo. transcription factors (TFs) miRNAs associated with predicted NetworkAnalyst database. total, 71 CoDEGs found be mainly involved cytokine–cytokine receptor interactions, tumor necrosis factor malaria signaling pathways. Animal experiments showed score, bronchoalveolar lavage fluid protein concentration, wet-to-dry weight ratio higher LPS group than those control group. Real-time polymerase chain reaction analysis indicated most such as colony-stimulating 2 (Csf2) overexpressed total 20 TFs including nuclear 1 (NRF1) two regulators genes. summary, Csf2 may serve diagnostic therapeutic target ALI.
